This has evoked an interest in these peptides as diagnostic\/prognostic tumor markers. If increased levels of beta-thymosins correspond to tumor malignancy, the question arises whether tumor growth inhibition induced by chemotherapeutic drugs would reduce their expression.          Two human breast cancer cell lines, the estrogen receptor(ER)-positive MCF-7 and the ER-negative MDA-MB231, were thus analyzed for the amount of beta-thymosin mRNAs by RNase protection assay and for the respective peptide levels by HPLC following different hormonal and drug treatments.          Both cell lines, growing in medium with 10% FCS, contain T beta(4) (400-500 fg\/cell) and Tbeta(10) (about 100 fg\/cell), but no T beta(15). Incubating MCF-7 cells with tamoxifen (1 microM) for 5 days resulted in about 80% growth inhibition and in reduction of intracellular T beta(4) and T beta(10) concentrations by about 40%. Levels of T beta(4) and T beta(10)-mRNA were reduced by about 60%. In contrast, cisplatin (2 microM) changed neither the peptide concentrations nor the mRNA levels of beta-thymosins, in spite of marked growth inhibition. In addition, no changes in beta-thymosin expression were observed in MDA-MB231 cells treated with either drug. MCF-7 cells maintained in estrogen-poor medium (10% horse serum) or stimulated to grow with estradiol (1 nM) had Tbeta(4) and T beta(10) concentrations reduced by about 30%, but changes in T beta(4)- and T beta(10)-mRNA levels did not correspond to those of the peptide.          Expression of T beta(4) and T beta(10) mRNAs and their peptides is differentially regulated and does not correlate with growth. Instead, reduced beta-thymosin expression may be linked to more intensive TRITC-phalloidin staining of F-actin lining the membrane at sites of intimate cell-cell contacts, while increased beta-thymosin levels appear in cells with more extensive substrate adhesion. This suggests that beta-thymosins play a role in cell surface dynamics.<\/p>\n<hr \/>\n<h2 id=\"Abs1\" class=\"c-article-section__title js-section-title js-c-reading-companion-sections-item\">\u5316\u7597\u836f\u7269\u6539\u53d8\u4eba\u4e73\u817a\u764c\u7ec6\u80de\u808c\u52a8\u86cb\u767d\u9aa8\u67b6\u7ed3\u6784\u548c\u03b2-\u80f8\u817a\u7d20\u7684\u8868\u8fbe\u3002<\/h2>\n<hr \/>\n<p>\u03b2-\u80f8\u817a\u7d20\u540c\u79cd\u578bT\u03b2(4)\u3001T\u03b2(10)\u548cT(15)\u7684\u9ad8\u8868\u8fbe\u4f3c\u4e4e\u4e0e\u4eba\u7c7b\u80bf\u7624\u7ec6\u80de(\u5305\u62ec\u4e73\u817a\u764c\u7ec6\u80de)\u7684\u6076\u6027\u8868\u578b\u8868\u73b0\u6709\u5173\u3002\u8fd9\u5f15\u8d77\u4e86\u4eba\u4eec\u5bf9\u8fd9\u4e9b\u80bd\u4f5c\u4e3a\u8bca\u65ad\/\u9884\u540e\u80bf\u7624\u6807\u5fd7\u7269\u7684\u5174\u8da3\u3002\u5982\u679c\u03b2-\u80f8\u817a\u7d20\u6c34\u5e73\u7684\u589e\u52a0\u5bf9\u5e94\u4e8e\u80bf\u7624\u6076\u6027\u7a0b\u5ea6\uff0c\u90a3\u4e48\u5316\u7597\u836f\u7269\u8bf1\u5bfc\u7684\u80bf\u7624\u751f\u957f\u6291\u5236\u662f\u5426\u4f1a\u964d\u4f4e\u5b83\u4eec\u7684\u8868\u8fbe\u5c31\u6210\u4e86\u95ee\u9898\u3002\u56e0\u6b64\uff0c\u5728\u4e0d\u540c\u7684\u6fc0\u7d20\u548c\u836f\u7269\u5904\u7406\u540e\uff0c\u901a\u8fc7RNase\u4fdd\u62a4\u8bd5\u9a8c\u5206\u6790\u4e86\u4e24\u79cd\u4eba\u4e73\u817a\u764c\u7ec6\u80de\u7cfb\uff0c\u96cc\u6fc0\u7d20\u53d7\u4f53(er)\u9633\u6027\u7684MCF-7\u548cER\u9634\u6027\u7684MDA-MB231\u7684\u03b2-\u80f8\u817a\u7d20mRNAs\u7684\u91cf\uff0c\u5e76\u901a\u8fc7HPLC\u5206\u6790\u4e86\u5404\u81ea\u7684\u80bd\u6c34\u5e73\u3002\u5728\u542b10% FCS\u7684\u57f9\u517b\u57fa\u4e2d\u751f\u957f\u7684\u4e24\u79cd\u7ec6\u80de\u7cfb\u90fd\u542b\u6709T\u03b2(4)(400-500 fg\/\u7ec6\u80de)\u548cT\u03b2(10)(\u7ea6100 fg\/\u7ec6\u80de)\uff0c\u4f46\u4e0d\u542bT\u03b2(15)\u3002\u5c06MCF-7\u7ec6\u80de\u4e0e\u4ed6\u83ab\u6614\u82ac(1 \u03bcm)\u4e00\u8d77\u5b75\u80b25\u5929\uff0c\u5bfc\u81f4\u7ea680%\u7684\u751f\u957f\u6291\u5236\u548c\u7ec6\u80de\u5185T\u03b2(4)\u548cT\u03b2(10)\u6d53\u5ea6\u964d\u4f4e\u7ea640%\u3002T\u03b2(4)\u548cT\u03b2(10)-mRNA\u7684\u6c34\u5e73\u964d\u4f4e\u4e86\u7ea660%\u3002\u76f8\u53cd\uff0c\u5c3d\u7ba1\u6709\u660e\u663e\u7684\u751f\u957f\u6291\u5236\uff0c\u987a\u94c2(2\u5fae\u6469\u5c14)\u65e2\u4e0d\u6539\u53d8\u03b2-\u80f8\u817a\u7d20\u7684\u80bd\u6d53\u5ea6\u4e5f\u4e0d\u6539\u53d8\u5176mRNA\u6c34\u5e73\u3002\u6b64\u5916\uff0c\u5728\u7528\u4efb\u4e00\u79cd\u836f\u7269\u5904\u7406\u7684MDA-MB231\u7ec6\u80de\u4e2d\u6ca1\u6709\u89c2\u5bdf\u5230\u03b2-\u80f8\u817a\u7d20\u8868\u8fbe\u7684\u53d8\u5316\u3002\u7ef4\u6301\u5728\u96cc\u6fc0\u7d20\u7f3a\u4e4f\u7684\u57f9\u517b\u57fa(10%\u9a6c\u8840\u6e05)\u4e2d\u6216\u7528\u96cc\u4e8c\u9187(1 nM)\u523a\u6fc0\u751f\u957f\u7684MCF-7\u7ec6\u80de\u7684T\u03b2(4)\u548cT\u03b2(10)\u6d53\u5ea6\u964d\u4f4e\u4e86\u7ea630%\uff0c\u4f46T\u03b2(4)-\u548cT\u03b2(10)-mRNA\u6c34\u5e73\u7684\u53d8\u5316\u4e0e\u80bd\u7684\u53d8\u5316\u4e0d\u4e00\u81f4\u3002T\u03b2(4)\u548cT\u03b2(10)mrna\u53ca\u5176\u80bd\u7684\u8868\u8fbe\u53d7\u5230\u4e0d\u540c\u7a0b\u5ea6\u7684\u8c03\u8282\uff0c\u4e0e\u751f\u957f\u65e0\u5173\u3002\u76f8\u53cd\uff0c\u03b2-\u80f8\u817a\u7d20\u8868\u8fbe\u7684\u51cf\u5c11\u53ef\u80fd\u4e0e\u7ec6\u80de\u95f4\u7d27\u5bc6\u63a5\u89e6\u90e8\u4f4d\u7684\u819c\u5185\u5c42F-\u808c\u52a8\u86cb\u767d\u7684\u66f4\u5f3aTRITC-phalloidin\u67d3\u8272\u6709\u5173\uff0c\u800c\u03b2-\u80f8\u817a\u7d20\u6c34\u5e73\u7684\u589e\u52a0\u51fa\u73b0\u5728\u5177\u6709\u66f4\u5e7f\u6cdb\u5e95\u7269\u7c98\u9644\u7684\u7ec6\u80de\u4e2d\u3002\u8fd9\u8868\u660e\u03b2-\u80f8\u817a\u7d20\u5728\u7ec6\u80de\u8868\u9762\u52a8\u529b\u5b66\u4e2d\u8d77\u4f5c\u7528\u3002<\/p>\n<\/div>\n","protected":false},"excerpt":{"rendered":"<p>Research
