\u7684 Nanopore \u571f\u58e4\u6570\u636e\u4e0a\uff0cnanoMDBG \u7528 <strong>125 GB \u5185\u5b58\u30016 \u5929<\/strong>\u5b8c\u6210\u7ec4\u88c5\uff1bmetaFlye \u5728\u540c\u4e00\u4efb\u52a1\u4e2d\u9700\u8981 <strong>753 GB \u5185\u5b58\u300128 \u5929<\/strong>\u3002<\/p>\n<p>\u8fd9\u5e76\u4e0d\u662f\u8bf4 125 GB \u53ef\u4ee5\u968f\u4fbf\u627e\u4e00\u53f0\u7535\u8111\u8dd1\u3002400 Gbp \u539f\u59cb reads\u3001\u7ea0\u9519\u4e2d\u95f4\u6587\u4ef6\u3001\u7ec4\u88c5\u56fe\u3001reads \u56de\u8d34 BAM \u548c\u540e\u7eed\u5206\u7bb1\u4ecd\u4f1a\u5360\u7528\u5927\u91cf CPU\u3001\u5185\u5b58\u4e0e\u78c1\u76d8\u3002\u771f\u6b63\u7a33\u59a5\u7684\u505a\u6cd5\uff0c\u662f\u5148\u6309\u6d4b\u5e8f\u7c7b\u578b\u548c\u6570\u636e\u590d\u6742\u5ea6\u9009 10%\u3002\u8fd9\u8bf4\u660e\u540e\u7eed\u56fe\u6784\u5efa\u548c\u89e3\u6790\u4ecd\u662f\u957f\u4efb\u52a1\uff0c\u4e0d\u80fd\u770b\u5230\u7ea0\u9519\u7ed3\u675f\u5c31\u5224\u65ad\u201c\u5feb\u8dd1\u5b8c\u4e86\u201d\u3002\u8c03\u5ea6\u65f6\u5e94\u4e3a\u6574\u4e2a \u538b\u7f29\u6587\u4ef6\u53ef\u80fd\u5bf9\u5e94\u4e0d\u540c\u603b\u78b1\u57fa\u91cf\uff1b\u571f\u58e4\u3001\u80a0\u9053\u548c mock community bases\u3001read N50\u3001\u8d28\u91cf\u5206\u5e03\u548c\u6837\u672c\u7c7b\u578b\uff0c\u800c\u4e0d\u662f\u53ea\u62a5\u201c\u4e00\u4e2a Nanopore \u6837\u672c\u201d\u3002<\/p>\n<p>\u6b63\u5f0f\u5168\u91cf\u7ec4\u88c5\u524d\uff0c\u53ef\u4ee5\u5bf9\u4ee3\u8868\u6027 reads \u5b50\u96c6\u505a\u73af\u5883\u9a8c\u6536\uff0c\u786e\u8ba4\u547d\u4ee4\u3001\u8f93\u51fa\u76ee\u5f55\u3001\u7248\u672c\u548c\u76d1\u63a7\u90fd\u6b63\u5e38\u3002\u4f46\u5b50\u96c6\u8fd0\u884c\u65f6\u95f4\u4e0e\u5185\u5b58\u4e0d\u80fd\u7b80\u5355\u7ebf\u6027\u653e\u5927\u5230\u5168\u91cf\uff1a\u4f4e\u4e30\u5ea6\u7269\u79cd\u3001\u590d\u6742\u5206\u652f\u548c\u957f\u91cd\u590d\u53ef\u80fd\u53ea\u5728\u6570\u636e\u589e\u5927\u540e\u51fa\u73b0\u3002\u57fa\u7ebf\u7684\u4f5c\u7528\u662f\u53d1\u73b0\u73af\u5883\u4e0e ONT\uff0c\u4f46\u8f93\u5165\u6a21\u5f0f\u4e0d\u540c\u3002Nanopore R10 simplex \u4f7f\u7528 <code>--in-ont<\/code>\uff0cPacBio HiFi \u4f7f\u7528 <code>--in-hifi<\/code>\u3002\u4e00\u4e2a\u53ef\u590d\u73b0\u7684 ONT \u8d77\u70b9\u5982\u4e0b\uff1a<\/p>\n<pre \u603b\u78b1\u57fa\u6570\u3001\u957f\u5ea6\u5206\u5e03\u3001\u5e73\u5747\u8d28\u91cf\u3001\u8fc7\u6ee4\u9608\u503c\u3001\u7ebf\u7a0b\u6570\u548c\u5de5\u4f5c\u76ee\u5f55\u3002\u8bba\u6587\u57fa\u51c6\u4f7f\u7528\u7684\u662f v1.2\uff1b\u5b98\u65b9\u4ed3\u5e93\u5728 2026 \u5e74 5 \u6708\u53d1\u5e03 v1.4\uff0c\u5e76\u8bf4\u660e\u6709\u8fd0\u884c\u65f6\u95f4\u3001\u5185\u5b58\u548c polishing \u66f4\u65b0\u3002\u65b0\u7248\u672c\u53ef\u80fd\u66f4\u5feb\uff0c\u4f46\u4e0d\u80fd\u76f4\u63a5\u628a v1.2 \u7684\u6570\u5b57\u5f53\u4f5c v1.4 \u6216\u81ea\u5df1\u6837\u672c\u7684\u4fdd\u8bc1\u3002<\/p>\n<p>\u8f93\u5165\u8d28\u63a7\u8981\u907f\u514d\u4e24\u4e2a\u6781\u7aef\u3002\u5b8c\u5168\u4e0d\u770b adapters\u3001\u77ed reads \u548c\u4f4e\u8d28\u91cf\u5c3e\u90e8\uff0c\u4f1a\u628a\u65e0\u6548\u5e8f\u5217\u5e26\u5165\u7ea0\u9519\uff1b\u8fc7\u5ea6\u8fc7\u6ee4\u53c8\u53ef\u80fd\u635f\u5931\u4f4e\u4e30\u5ea6\u57fa\u56e0\u7ec4\u8986\u76d6\u3002\u81f3\u5c11\u4fdd\u5b58\u8fc7\u6ee4\u524d\u540e reads \u6570\u3001\u603b bases\u3001\u957f\u5ea6\u4e0e\u8d28\u91cf\u5206\u5e03\uff0c\u5e76\u628a\u8fc7\u6ee4\u547d\u4ee4\u5199\u5165\u65e5\u5fd7\u3002\u6d89\u53ca\u4eba\u6e90\u6216\u5bbf\u4e3b\u6837\u672c\u65f6\uff0c\u8fd8\u8981\u6839\u636e\u4f26\u7406\u548c\u5206\u6790\u76ee\u7684\u51b3\u5b9a\u5bbf\u4e3b\u8fc7\u6ee4\u53d1\u751f\u5728\u7ec4\u88c5\u524d\u8fd8\u662f\u7ed3\u679c\u9636\u6bb5\uff0c\u4e0d\u80fd\u9ed8\u8ba4\u628a\u654f\u611f reads \u5e26\u5165\u5171\u4eab\u73af\u5883\u3002<\/p>\n<p>\u73af\u5883\u4e5f\u5e94\u56fa\u5b9a\uff1a\u4fdd\u5b58 metaMDBG\u3001minimap2\u3001samtools \u7248\u672c\u53ca Conda\/\u5bb9\u5668\u6e05\u5355\uff1b\u540c\u4e00\u9879\u76ee\u4e0d\u8981\u4e2d\u9014\u5347\u7ea7 assembler\u3002<\/p>\n<p>ONT R9\u3001\u65e9\u671f\u9ad8\u9519\u8bef\u7387 reads\u3001\u8d85\u957f reads \u4e0e R10 simplex \u7684\u8bef\u5dee\u7ed3\u6784\u4e0d\u540c\u3002\u82e5\u6570\u636e\u7c7b\u578b\u4e0d\u5339\u914d\uff0c\u7b97\u6cd5\u4f18\u52bf\u53ef\u80fd\u6d88\u5931\u3002\u5bf9\u4e8e HiFi\uff0c\u5e94\u8be5\u8d70 metaMDBG \u7684 HiFi \u6a21\u5f0f\uff0c\u5e76\u4e0e hifiasm-meta\u3001metaFlye \u7684\u7ed3\u679c\u548c\u7814\u7a76\u76ee\u6807\u4e00\u8d77\u8bc4\u4f30\uff0c\u800c\u4e0d\u662f\u56e0\u4e3a nanoMDBG \u5728 ONT \u57fa\u51c6\u4e2d\u66f4\u7701\u8d44\u6e90\u5c31\u8de8\u5e73\u53f0\u5957\u7528\u3002<\/p>\n<h2 style=\"margin-top:34px;color:#12314a;\">\u670d\u52a1\u5668\u89c4\u5212\u8981\u8986\u76d6\u201c\u7ec4\u88c5\u4e4b\u540e\u201d<\/h2>\n<p>\u539f\u59cb FASTQ \u53ea\u662f\u7b2c\u4e00\u4efd\u6570\u636e\u3002\u7ec4\u88c5\u76ee\u5f55\u3001\u7ea0\u9519 reads\u3001\u4e34\u65f6\u56fe\u3001contigs\u3001GFA\u3001reads-to-contigs BAM\u3001coverage \u8868\u548c MAG \u5206\u7bb1\u7ed3\u679c\u53ef\u80fd\u540c\u65f6\u5b58\u5728\u3002\u5c24\u5176\u662f reads \u56de\u8d34\uff0c\u5982\u679c\u5148\u5199\u5de8\u5927 SAM \u518d\u6392\u5e8f\uff0c\u4f1a\u4ea7\u751f\u4e0d\u5fc5\u8981\u7684 I\/O \u5cf0\u503c\u3002\u66f4\u5408\u7406\u7684\u662f\u6d41\u5f0f\u4f20\u7ed9 samtools\uff1a<\/p>\n<pre SSD\/NVMe\uff1b\u4e34\u65f6\u6587\u4ef6\u53ef\u7528\u672c\u5730 scratch\uff0c\u4f46 checkpoint \u548c\u5173\u952e\u4e2d\u95f4\u6587\u4ef6\u4e0d\u80fd\u53ea\u653e\u4f1a\u81ea\u52a8\u6e05\u7406\u7684\u4e34\u65f6\u76d8\u3002\u5171\u4eab\u6587\u4ef6\u7cfb\u7edf\u62e5\u5835\u65f6\uff0cCPU \u4f7f\u7528\u7387\u4f1a\u4e0b\u964d\uff0c\u4efb\u52a1\u770b\u4f3c\u201c\u8fd8\u5728\u8dd1\u201d\u5374\u957f\u671f\u7b49\u5f85 I\/O\u3002\u5148\u76d1\u63a7\u8bfb\u5199\u5e26\u5bbd\u548c\u961f\u5217\uff0c\u518d\u51b3\u5b9a\u589e\u52a0\u7ebf\u7a0b\u3002<\/p>\n<p>\u5efa\u8bae\u5728\u57fa\u7ebf\u4efb\u52a1\u5916\u5c42\u4f7f\u7528 <code>\/usr\/bin\/time -v<\/code> \u8bb0\u5f55\u5cf0\u503c\u5185\u5b58\u4e0e\u5899\u949f\u65f6\u95f4\uff0c\u540c\u65f6\u5b9a\u671f\u91c7\u96c6 <code>iostat<\/code>\u3001\u5269\u4f59\u7a7a\u95f4\u548c\u8f93\u51fa\u76ee\u5f55\u589e\u957f\u3002\u53ea\u770b CPU \u767e\u5206\u6bd4\u65e0\u6cd5\u533a\u5206\u8ba1\u7b97\u3001I\/O \u7b49\u5f85\u548c\u5185\u5b58\u6362\u9875\uff1b\u5982\u679c swap \u5f00\u59cb\u6301\u7eed\u589e\u957f\uff0c\u5e94\u5148\u505c\u6b62\u653e\u5927\u5168\u91cf\u4efb\u52a1\u3002\u8f93\u51fa\u5b8c\u6210\u540e\u4e3a\u539f\u59cb reads\u3001contigs\u3001\u914d\u7f6e\u548c\u5173\u952e\u62a5\u544a\u751f\u6210\u6821\u9a8c\u503c\uff0c\u8fc1\u79fb\u5230\u5f52\u6863\u76d8\u540e\u518d\u6e05\u7406\u5de5\u4f5c\u76ee\u5f55\u3002<\/p>\n<p>\u591a\u6837\u672c\u5e76\u53d1\u4e5f\u8981\u6309\u5cf0\u503c\u7b97\u3002\u4e24\u4efd\u5404\u9700 125 GB \u7684\u571f\u58e4\u4efb\u52a1\u540c\u65f6\u8fd0\u884c\uff0c\u4e0d\u80fd\u53ea\u7533\u8bf7\u4e00\u53f0 256 GB \u8282\u70b9\u540e\u8ba9\u7cfb\u7edf\u81ea\u7531\u7ade\u4e89\uff1b\u8fd8\u8981\u7ed9\u64cd\u4f5c\u7cfb\u7edf\u3001\u6587\u4ef6\u7f13\u5b58\u3001\u56de\u8d34\u548c\u4e0b\u6e38\u6d41\u7a0b\u7559\u4f59\u91cf\u3002\u957f\u8fbe\u6570\u5929\u7684\u4efb\u52a1\u5e94\u8bbe\u7f6e\u65e5\u5fd7\u3001\u78c1\u76d8\u544a\u8b66\u548c\u5931\u8d25\u6062\u590d\u68c0\u67e5\uff0c\u907f\u514d\u5728\u7b2c HiFi \u5b8f\u57fa\u56e0\u7ec4\u548c\u591a\u79cd assembler \u7684\u590d\u67e5\u6307\u51fa\uff0c\u957f\u8bfb\u957f\u7ec4\u88c5\u53ef\u51fa\u73b0\u6bcf 1 \u4ebf bp \u8d85\u8fc7 <strong>40 \u4e2a\u9519\u8bef<\/strong>\uff0c\u5305\u62ec\u5d4c\u5408\u3001\u8fc7\u65e9\u73af\u5316\u3001\u5355\u500d\u578b\u9519\u8bef\u3001\u91cd\u590d\u548c phantom sequence\u3002\u8bba\u6587\u7279\u522b\u63d0\u9192\uff1a\u62a5\u544a\u4e3a circular \u7684 contig \u4e5f\u53ef\u80fd\u7f3a\u5931\u6838\u5fc3\u57fa\u56e0\u7ec4\u7247\u6bb5\u3002<\/p>\n<p>\u8d28\u91cf\u9a8c\u6536\u6700\u597d\u5206\u4e09\u5c42\u3002\u7b2c\u4e00\u5c42\u770b reads \u56de\u8d34\u7387\u3001split alignment\u3001\u96f6\u8986\u76d6\u533a\u548c\u8986\u76d6\u7a81\u53d8\uff0c\u5224\u65ad contig \u662f\u5426\u6709\u76f4\u63a5 read \u652f\u6301\uff1b\u7b2c\u4e8c\u5c42\u770b CheckM2\u3001GUNC \u7b49\u5b8c\u6574\u6027\u4e0e\u6c61\u67d3\u6307\u6807\uff0c\u5224\u65ad MAG \u662f\u5426\u50cf\u4e00\u4e2a\u4e00\u81f4\u7684\u57fa\u56e0\u7ec4\uff1b\u7b2c\u4e09\u5c42\u770b GTDB \u5206\u7c7b\u3001\u6838\u5fc3\u4ee3\u8c22\u4e0e\u86cb\u767d\u5b8c\u6574\u6027\uff0c\u5224\u65ad\u7ed3\u679c\u662f\u5426\u7b26\u5408\u751f\u7269\u5b66\u80cc\u666f\u3002\u4efb\u4f55\u5355\u4e00\u6307\u6807\u90fd\u4e0d\u80fd\u66ff\u4ee3\u53e6\u5916\u4e24\u5c42\u3002<\/p>\n<p>nanoMDBG \u7684 2026 \u8bba\u6587\u540c\u6837\u663e\u793a\u65b9\u6cd5\u4e4b\u95f4\u6709\u53d6\u820d\u3002ONT \u957f contig \u7684\u6c61\u67d3\u6bd4\u4f8b\u4e2d\u4f4d\u6570\u90fd\u8f83\u4f4e\uff0c\u4f46 metaFlye \u7684\u6c61\u67d3\u957f contig \u4e3a 2\/326\uff0cnanoMDBG \u4e3a 13\/1006\uff1b\u53e6\u4e00\u65b9\u9762\uff0cnanoMDBG \u7684 clipping events\u3001\u96f6\u8986\u76d6\u533a\u57df\u548c\u5168\u957f\u86cb\u767d\u6307\u6807\u5728\u591a\u4e2a\u6570\u636e\u96c6\u4e0a\u4f18\u4e8e metaFlye\u3002\u7ed3\u8bba\u4e0d\u662f\u201c\u4e00\u4e2a\u5de5\u5177\u6c38\u8fdc\u6700\u597d\u201d\uff0c\u800c\u662f\u7ec4\u88c5\u540e\u5fc5\u987b\u628a reads \u56de\u8d34\uff0c\u68c0\u67e5 clipping\u3001\u96f6\u8986\u76d6\u3001\u6c61\u67d3\u3001\u5b8c\u6574\u6027\u548c\u57fa\u56e0\u9884\u6d4b\u3002<\/p>\n<p>\u5706\u5f62 contig \u4e5f\u4e0d\u80fd\u76f4\u63a5\u7b49\u540c\u4e8e\u5b8c\u6574\u67d3\u8272\u4f53\u6216\u8d28\u7c92\u3002\u81f3\u5c11\u8981\u6838\u5bf9\u8986\u76d6\u662f\u5426\u8fde\u7eed\u3001\u672b\u7aef\u662f\u5426\u6709\u5f02\u5e38 reads split\u3001\u5355\u62f7\u8d1d\u6838\u5fc3\u57fa\u56e0\u662f\u5426\u5b8c\u6574\uff0c\u4ee5\u53ca\u8fd1\u7f18\u53c2\u8003\u4e2d\u5e94\u5b58\u5728\u7684\u6838\u5fc3\u529f\u80fd\u662f\u5426\u7f3a\u5931\u3002\u5bf9\u5173\u952e MAG\uff0c\u5efa\u8bae\u4fdd\u7559\u591a assembler \u7ed3\u679c\u6216\u81f3\u5c11\u505a\u9488\u5bf9\u6027\u590d\u6838\u3002<\/p>\n<p>\u5982\u679c\u9879\u76ee\u76ee\u6807\u662f\u627e\u75c5\u6bd2\u3001\u8d28\u7c92\u6216\u7279\u5b9a\u4f4e\u4e30\u5ea6\u7269\u79cd\uff0c\u8bc4\u4ef7\u6807\u51c6\u8fd8\u8981\u8c03\u6574\u3002\u8ffd\u6c42\u6700\u957f contig \u8d28\u63a7\uff0c\u53ef\u4ee5\u51c6\u5907\u6d4b\u5e8f\u5e73\u53f0\u4e0e chemistry\u3001reads \u603b\u91cf\u548c\u8d28\u91cf\u3001\u6837\u672c\u7c7b\u578b\u3001\u73b0\u6709\u670d\u52a1\u5668\u548c\u76ee\u6807\u4ea4\u4ed8\u3002\u4e91\u751f\u4fe1\u53ef\u5148\u505a\u5de5\u5177\u9002\u914d\u4e0e\u5c0f\u89c4\u6a21\u57fa\u7ebf\uff0c\u518d\u786e\u8ba4\u79df\u7528\u73af\u5883\u81ea\u884c\u8fd0\u884c\u3001\u670d\u52a1\u5668\u90e8\u7f72\u4ea4\u63a5\u6216\u59d4\u6258\u5206\u6790\u7684\u8303\u56f4\u3002<\/p>\n<p><a
